You can force release a tuner, even if another application has set a key and locked a tuner. In general, this type of functionality exists to free a tuner that a crashed or unresponsive application locked. It is not always successful, but it does indeed work. (The reason I say not always successful is that some software will attempt to grab a tuner immediately upon freeing; this is especially the case with SiliconDust's own DVR software. There is no easy way to stop a recording-in-progress with their software, short of literally unplugging the tuner.)
In any case, the commands to release a tuner are:
hdhomerun_config FFFFFFFF set /tunerX/lockkey force
Change FFFFFFFF to your tuner's DeviceID, otherwise the first discovered tuner will be used. (If you have only a single tuner, that usually doesn't matter. But, if you have multiple tuners, the first discovered tuner may not be the device you want.) Also, change tunerX to the proper tuner. (Remember, tuners are 0-indexed, so the first tuner is tuner0.)

(After unlocking a tuner, you probably will want to clear it. I don't believe that unlocking a tuner does this step as well, so to ensure the tuner is ready for release, you should free it after unlocking:
hdhomerun_config FFFFFFFF set /tunerX/channel none
