I’m able to receive around 408 Plex Channels at of the 600.
Is that about right?

408 channels is what I get using the "Gracenote" playlist. There's another 218 in the "No Gracenote" playlist. So, using the "two-source method" I get about 626 channels.
How do you add both playlist into channels?

I'm not really good at explaining how to do stuff. But, I'll give it a shot...
I'm assuming from your post that you probably have the first Custom Source added.
For instance, this is my "Plex with Gracenote EPG" source, which has the 408 channels:

Of course, you'll have to replace my "192.168.1.100:7776" with your own DVR IP address and Plex Port #.

This additional source should give you the other 218 channels. They will have minimal guide data.

It is possible to have a single combined source with all 626 channels.

As I understand, it's preferable to separate the two sources based upon the Gracenote data availability.
I'm sure that there's someone more knowledgeable than I, who can help explain why that is so.
It comes down to matching. Channels uses Gracenote as the core backend of metadata. XML metadata doesn't match this, so you end up with two "versions" of the same program. I explain it in more detail and clarity here:
@sdarnell63, due to this, you want the Gracenote data whenever you can get it. Also, there's two weeks worth instead of a few days. Further, you're already paying for Gracenote data with your Channels subscription, so always better to use what you've invested in!
Now, if you or anyone is going to be adding multiple playlists and want an easier way to manage them with Channels, then I'll just plug myself here with this:

You need to use the XMLTV URL in your source settings to get the Plex guide data for those non-Gracenote channels.
